What each ingredient does

Ripe strawberries
Supply the juice, flavour and aroma of the drink.
Powdered loaf sugar
Draws water out of the fruit and dissolves into the juice, adding sweetness.

The step that matters

Letting the mixture stand for 12 hours. Sugar draws liquid out of the strawberry cells by osmosis, so the juice forms over time. Standing too briefly yields less juice and a weaker flavour, so the full period matters.
